Nirogacestat’s Mechanism of Action to Treat Desmoid Tumors

Mary Smith, PhD, senior vice president of clinical research and development at SpringWorks Therapeutics explains the how nirogacestat works to treat individuals with Desmoid tumors.

Desmoid tumors are benign tumors that can grow into surrounding healthy tissues, including joints, muscle and viscera. These rare tumors can cause significant pain, internal bleeding, and reduced mobility.

As Smith explains in this video, nirogacestat is an oral, selective, small molecule, gamma secretase inhibitor. Gamma secretase cleaves multiple transmembrane protein complexes, including Notch, which may contribute to desmoid tumor growth. The drug is currently under investigation in a Phase 3 clinical trial involving adults and a Phase 2 clinical trial involving children with Desmoid tumors.
